How do i use radioactive isotope's to find out whether the oxygen in CO2 comes from sugar or oxygen gas?

You would have to label plant photosynthetic product with the radioactive oxygen, then feed the material to the primary consumer. When the sugar was broken down, you would be able to see whether the oxygen was labeled. If it was, it obviously came from the plant product.
